The PathHunter DRD4 eXpress kit provides an easy-to-use cell based assay to study drug candidates (small molecules or biologics) targeting GPCRs. This kit assesses ligand (e.g. dopamine) based activation of DRD4 GPCR activity via detection of β-arrestin recruitment. The included cell line overexpresses ProLink-tagged DRD4 and EA-tagged β-Arrestin-2. Activation of DRD4 stimulates the recruitment of β-Arrestin-2 and produces EFC signal. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. The PathHunter C5AR1 Activated GPCR internalization eXpress kit provides an easy to-use cell based assay to study drug candidates (small molecules or biologics) affecting GPCR activation and internalization. This kit uses EFC technology to measure ligand-induced internalization of C5AR1 via detection of localization to the early endosome. The included cell line overexpresses EA-tagged β-Arrestin-2 as well as a ProLink-tagged early endosome protein and untagged C5AR1. Upon ligand-induced receptor activation, β-Arrestin-2 is recruited to the activated GPCR and the resulting C5AR1-arrestin complex is internalized to the early endosome, leading to production of EFC signal. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. The cells overexpress wild type rDRD2L, a naturally Gi/Go coupled GPCR. Upon activation of rDRD2L, the decrease in levels of intracellular cAMP leads to a decreased signal from EFC when the cAMP detection kit is applied. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates.
